Description

A Sandwich ELISA developed by Assay Genie for the quantitative measurement of human Disintegrin and metalloproteinase domain-containing protein 17 (ADAM17), also known by synonyms including TNF-alpha convertase, TNF-alpha-converting enzyme, CD156b, CSVP, TACE and Snake venom-like protease. The assay uses a colorimetric detection method and is validated for serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids. Specificity has been defined for natural and recombinant human ADAM17.

According to the manufacturer's specifications, the kit reports a detection range of 0.312–20 ng/mL and a sensitivity of 0.11 ng/mL. Precision is given as an intra-assay CV of 5.2% and an inter-assay CV of 10.3%. Linearity was assessed in serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16, with reported values spanning roughly 80–120% depending on matrix. Spike-and-recovery gave an average of 103% (range 97–109%) for serum and 105% (range 99–111%) for plasma.

ADAM17 is a single-pass type I membrane protein with disintegrin and metalloprotease activity. It cleaves the membrane-bound precursor of TNF-alpha to its mature soluble form and mediates shedding of cell-surface proteins including p75 TNF-receptor, interleukin 1 receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1 and the amyloid precursor protein. It also acts as an activator of the Notch pathway and contributes to the proteolytic processing of ACE2. The associated UniProt entry is P78536. The product is grouped within the Neuroscience research area and is supplied for research use only.
