Description

A Sandwich ELISA kit from Assay Genie, configured as a double-antibody immunoassay, enables quantitative detection of human DEXI (Dexamethasone-Induced Protein) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay is read colorimetrically at 450 nm and is intended for cell biology research. Alternate names for the target include MYLE and Protein MYLE (Uniprot O95424).

Reported sensitivity is 0.469 ng/mL across a working range of 0.781–50 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions (n=5) was 85–104% / 83–100% / 81–100% for serum, 85–103% / 85–100% / 82–95% for EDTA plasma, and 93–103% / 88–97% / 90–104% for heparin plasma. Spike-and-recovery averaged 92% in serum (range 94–103%, n=5), 99% in EDTA plasma (range 85–104%, n=5), and 92% in heparin plasma (range 85–98%, n=5).

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks on the pre-coated plate for a 90-minute incubation at 37°C, binding the biotinylated detection antibody for 60 minutes at 37°C, adding HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm immediately after adding stop solution. The kit should be stored at 2–8°C for up to 12 months.
