Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of human Delta-1-pyrroline-5-carboxylate dehydrogenase, mitochondrial (ALDH4A1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay is reactive with human samples and uses a colorimetric detection method, providing a reliable tool for research into proline metabolism and related disease pathways.

Performance characteristics include a detection range of 0.156–10 ng/mL and a sensitivity of 0.093 ng/mL. Precision is supported by an intra-assay CV of 6.1% and an inter-assay CV of 10.2%. Linearity has been validated across serum, EDTA plasma and heparin plasma at dilutions from 1:2 to 1:16, with recovery averaging 96% (range 90–102%) for serum and 98% (range 92–104%) for plasma. The assay specifically detects natural and recombinant human ALDH4A1, a mitochondrial matrix enzyme that catalyses the irreversible conversion of delta-1-pyrroline-5-carboxylate (P5C) to glutamate, linking the urea and tricarboxylic acid cycles; the preferred substrate is glutamic gamma-semialdehyde.

Dysregulation of ALDH4A1 has been associated with cancer, cardiovascular diseases and neurodegenerative disorders, and defects in the enzyme are linked to hyperprolinemia type II. The protein functions as a homodimer within the mitochondrion matrix and belongs to the aldehyde dehydrogenase family. For research use only.
