Paneth cells positioned at the base of intestinal crypts of Lieberkühn secrete Defensin Alpha 5 (DEFa5), a 32-residue antimicrobial peptide that contributes to innate mucosal defense in the small intestine. By disrupting microbial membranes, DEFa5 limits bacterial, fungal, and viral colonization and helps shape the composition of the resident gut microbiota. Reduced or dysregulated DEFa5 secretion has been reported in Crohn's disease, enteric infection studies, and investigations of mucosal barrier integrity, making accurate quantification of this peptide valuable in mucosal immunology research.

The Assay Genie colorimetric kit enables quantitative measurement of human DEFa5 across diverse research matrices, including serum fractions, plasma samples, tissue homogenates, cell lysates, and clarified cell culture supernatant. The supplied 96-assay microtiter plate format supports cohort-scale screening, longitudinal sampling designs, and parallel processing of experimental and control specimens. Each kit contains pre-coated wells, a standard curve set of calibrators, detection reagent, wash concentrate, substrate, stop solution, and a full protocol booklet.

Reconstituted standard aliquots should be stored frozen at -20°C or below for working-stock stability, while working dilutions are prepared fresh before each run. Absorbance is read on a standard microplate reader, and DEFa5 concentrations in experimental samples are interpolated from the standard curve. The kit is configured for in vitro research use only and is not validated for diagnostic procedures.
