Description

Assay Genie offers a sandwich ELISA built on a double-antibody format for the quantitative measurement of human DEFA1 (Defensin Alpha 1, Neutrophil). The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples of human origin, and is supplied for research use only. Defensin alpha 1 is an antimicrobial peptide of the innate immune system that acts by disrupting microbial cell membranes and has been implicated in inflammatory conditions, making it a useful biomarker for immunology and infectious-disease research.

Assay sensitivity is quoted at 18.75 pg/mL, with a working range of 31.25-2000 pg/mL. Spike-and-recovery testing gave average recoveries of 92% in serum (range 88-100%), 90% in EDTA plasma (88-92%) and 97% in heparin plasma (88-104%), each at n=5. Linearity was assessed by serial dilution of spiked samples: serum (n=5) 87-103% at 1:2, 86-102% at 1:4 and 90-103% at 1:8; EDTA plasma (n=5) 85-98%, 84-101% and 83-99%; heparin plasma (n=5) 80-100%, 82-100% and 80-96%.

The protocol uses a biotin-labeled detection antibody followed by an HRP-streptavidin conjugate (SABC) and TMB substrate, with absorbance read at 450 nm after addition of stop solution. Supplied components include a dismountable ELISA microplate, lyophilized standard, concentrated biotin-labeled detection antibody, HRP-streptavidin conjugate (SABC), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer and a technical manual. Recommended storage is 2-8°C, under which the kit remains stable for 12 months.
