Quantitative measurement of Decay Accelerating Factor (DAF, also called CD55) in human specimens with this immunoassay from Assay Genie. DAF is a GPI-anchored complement regulatory protein found on circulating blood cells and many epithelia, where it accelerates the decay of C3 and C5 convertases to protect host tissue from autologous complement attack. Altered DAF expression is studied in paroxysmal nocturnal hemoglobinuria, autoimmune hemolysis, graft rejection, and tumor immune-evasion research.

The kit ships in a microplate format with pre-coated strips, lyophilized recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer concentrate, TMB substrate, and a stop vial. Reconstituted standards should be aliquoted and frozen to limit freeze-thaw cycles, and unopened kit components should be refrigerated at 4 degrees C until first use. The conjugate and substrate are light-sensitive and must be protected from light during incubations.

Compatible with cell culture supernatants, tissue lysates, and clarified specimens, the protocol accommodates the small sample volumes typical of immunology and complement research. The assay is for in vitro research use only and is not cleared for diagnostic applications. Bring the plate and all working solutions to room temperature before initiating the run, and read absorbance shortly after the stop step.
