Description

A Sandwich ELISA, Double Antibody assay from Assay Genie designed for the quantitative detection of human L-dopachrome tautomerase (DCT) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric detection method, with sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL, and is stored at 2-8°C for 12 months.

The assay procedure uses a pre-coated microplate with standards, samples and blanks loaded for primary incubation at 37°C for 90 minutes, followed by a biotin-labeled detection antibody incubated at 37°C for 60 minutes, HRP-Streptavidin (SABC) at 37°C for 30 minutes, TMB substrate development in the dark for 10-20 minutes, and absorbance read at 450 nm after stop solution is added. Linearity for serum (n=5) at 1:2, 1:4 and 1:8 dilutions is 82-100%, 81-100% and 90-98%; EDTA plasma (n=5) is 82-98%, 82-92% and 85-105%; and heparin plasma (n=5) is 85-100%, 87-101% and 82-99%. Recovery in serum (n=5) is 87-103% (average 95%), in EDTA plasma (n=5) is 85-100% (average 93%), and in heparin plasma (n=5) is 85-105% (average 90%).

DCT, also known as L-dopachrome tautomerase, TYRP2, TRP-2, TRP2, DT or L-dopachrome Delta-isomerase, is a tyrosinase-family enzyme involved in melanin biosynthesis and pigmentation. Synonyms listed in the source documentation include DCT, L-dopachrome tautomerase, TYRP2, DCT, DT, L-dopachrome Delta-isomerase, Tyrosinase-related protein 2, TRP-2 and TRP2, with UniProt reference P40126.
