Human Cytokeratin ELISA Kit

A Sandwich ELISA kit from Assay Genie developed for the quantitative measurement of cytokeratin in human samples. The assay uses a double-antibody sandwich format with colorimetric detection, supporting research applications in cell biology where cytokeratins serve as key structural proteins in epithelial cells.

Linearity at 1:2, 1:4 and 1:8 dilutions was 87-101%, 84-100% and 80-98% for serum; 89-105%, 92-101% and 82-99% for EDTA plasma; and 88-105%, 85-97% and 82-94% for heparin plasma. Spike recovery averaged 100% in serum (range 92-105%), 96% in EDTA plasma (range 89-105%) and 94% in heparin plasma (range 85-103%).

The protocol proceeds through reagent and TMB substrate equilibration at room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB substrate development in the dark for 10-20 minutes, and a stop reaction with absorbance measured at 450 nm.
