Human Cytokeratin 10/KRT10 ELISA Kit

From Assay Genie, this Sandwich ELISA is intended for the quantitative measurement of human Cytokeratin 10 (KRT10) in serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The assay uses a double-antibody sandwich format with colorimetric detection, and absorbance is read at 450 nm. The kit reports a sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL.

Cytokeratin 10 contributes to the structural integrity of epidermal skin cells, and altered KRT10 expression has been associated with skin disorders, supporting its use as a biomarker in dermatology and skin biology research. Validated performance includes serum recovery of 88–100% (average 97%), EDTA plasma recovery of 87–102% (average 95%) and heparin plasma recovery of 89–104% (average 94%). Linearity is reported as 82–97%, 88–102% and 80–91% for serum, 90–97%, 85–91% and 92–97% for EDTA plasma, and 90–102%, 88–105% and 84–100% for heparin plasma, across 1:2, 1:4 and 1:8 dilutions. Unopened kits are stored at 2–8°C for 12 months.

The procedure involves equilibrating reagents to room temperature, a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody step, a 30-minute HRP-Streptavidin binding step, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution is added. Kit components include a pre-coated dismountable microplate, lyophilized standard, 100X biotin-labelled antibody, 100X HRP-Streptavidin conjugate (SABC), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealers and a technical manual.
