COX1 is the largest catalytic core subunit of cytochrome c oxidase, the terminal complex (Complex IV) of the mitochondrial electron transport chain. Encoded by the mitochondrial genome (MT-CO1 gene), this transmembrane heme-copper protein contains the heme a and heme a3 centers that drive oxygen reduction and contribute to the proton gradient driving ATP synthesis. Quantitative measurement of COX1 expression is widely used in studies of mitochondrial biogenesis, oxidative phosphorylation efficiency, and the metabolic remodeling observed in cancer, neurodegeneration, and inherited mitochondrial disorders.

This Assay Genie immunoassay kit provides a colorimetric microplate workflow for quantifying human COX1 in tissue homogenates, cell lysates, and other body fluid preparations. The plate includes all reagents required to run the full 96 assays, supporting standard curve-based measurement in research laboratories. Samples and standards are processed through antibody-coated wells, with absorbance read on a standard microplate reader, making the workflow compatible with high-throughput mitochondrial profiling screens.

Intended for research use only, the kit supports investigation of mitochondrial content changes, mtDNA-encoded versus nuclear-encoded subunit regulation, and downstream functional assays in muscle, neural, and cardiac tissues where oxidative capacity is a key experimental readout.
