Human CYP3A5/Cytochrome P450 3A5 ELISA Kit

Designed for the in vitro quantitative determination of human CYP3A5, this sandwich ELISA from Assay Genie uses a double-antibody format with a colorimetric detection method. The kit is intended for research use only and is suited to studies of drug metabolism, pharmacogenomics, and personalized medicine, where CYP3A5 expression has been linked to differences in drug efficacy and toxicity. The assay has been validated for use with serum, plasma, cell culture supernatants, and other biological fluids.

In spike-and-recovery testing, serum showed an average recovery of 96% (range 88–103%, n=5), EDTA plasma 92% (range 85–101%, n=5), and UFH plasma 93% (range 86–103%, n=5). Linearity across serial dilutions was 87–102% (1:2), 87–98% (1:4), and 86–101% (1:8) for serum; 82–94%, 87–100%, and 86–100% for EDTA plasma; and 86–97%, 86–98%, and 81–100% for UFH plasma. Precision is reported as intra-assay CV <8% and inter-assay CV <10%. The kit should be stored at 4°C for 6 months.

According to UniProt entry P20815, CYP3A5 is a heme-thiolate monooxygenase that functions in an NADPH-dependent electron transport pathway in liver microsomes, oxidizing steroids, fatty acids, and xenobiotics. The protein is encoded on chromosome 7q21.1 and exists as two isoforms generated by alternative splicing.
