Description

Assay Genie offers a sandwich ELISA kit for the quantitative measurement of Cyclin D1 in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody format with colorimetric detection, and Cyclin D1 is described as a key protein that regulates cell cycle progression and is associated with the development of various diseases, including cancer and neurodegenerative disorders.

The kit has a stated sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL, and it should be stored at 2-8°C for 12 months. Reported linearity values include serum (n = 5) at 87-96% (1:2), 94-102% (1:4) and 86-98% (1:8); EDTA plasma (n = 5) at 90-103% (1:2), 87-92% (1:4) and 88-91% (1:8); and heparin plasma (n = 5) at 85-98% (1:2), 83-100% (1:4) and 82-100% (1:8). Reported recovery averages are 92% for serum (range 87-104%), 97% for EDTA plasma (range 86-103%) and 93% for heparin plasma (range 86-105%).

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, colour development with TMB substrate in the dark for 10-20 minutes, and a stop reaction read at 450 nm. Kit components include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-streptavidin conjugate SABC (100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual.
