Description

A high-quality sandwich ELISA from Assay Genie designed for the quantitative measurement of human CXCL5 (also known as ENA-78), a chemokine involved in inflammatory processes. The assay uses a double-antibody sandwich format with a colorimetric detection method, employing TMB substrate read at 450 nm.

Validated for human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, the kit offers a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. Linearity has been demonstrated at 1:2, 1:4, and 1:8 dilutions: serum 88–104%, EDTA plasma 82–101%, and heparin plasma 80–101%. Mean recovery across serum, EDTA plasma, and heparin plasma samples falls within 91–92%.

The protocol includes reagent equilibration, a 90-minute primary incubation at 37°C, biotinylated detection antibody binding for 60 minutes, HRP-Streptavidin incubation for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution. Kit components comprise a pre-coated microplate, lyophilised standard, biotin-labelled detection antibody, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer, and a technical manual. Most reagents are stored at 2–8°C; the microplate and standard are also suitable for longer-term storage at –20°C. CXCL5/ENA-78 attracts neutrophils to sites of infection or injury, and its dysregulation has been linked to inflammatory disorders such as arthritis, asthma, and chronic obstructive pulmonary disease, making this kit a useful tool for studying inflammation and developing potential therapies.
