The CTR target is encoded by the CALCR gene, located on human chromosome 7q21.3, and belongs to the GPCR family 2 (secretin-receptor subfamily) of seven-transmembrane receptors. Calcitonin binding triggers Gs-coupled activation of adenylyl cyclase, raising intracellular cAMP and inhibiting osteoclast-mediated bone resorption, while alternative splicing generates at least three receptor isoforms with tissue-specific expression patterns. The Assay Genie sandwich ELISA detects native and recombinant receptor protein and shows no significant cross-reactivity with structurally related analogues such as calcitonin gene-related peptide or amylin receptors.

After incubation with HRP-conjugated detector antibody and washing, the chromogenic substrate is added and the enzymatic reaction is stopped with sulphuric acid; absorbance is read at 450 nm on a standard microplate reader and interpolated against the supplied standard curve. Comparable sandwich-format CTR kits typically report sensitivity in the low ng/mL range (for example 0.094–10 ng/mL or 50–1000 pg/mL depending on configuration), with intra-assay CV below 10%, inter-assay CV below 15%, and spike-and-recovery between 85% and 115%. Researchers should consult the lot-specific Certificate of Analysis shipped with the kit for the exact standard concentrations, expected dynamic range, and validated dilution recommendations for each sample matrix.

Sample handling tips: collect serum or plasma using standard anticoagulant tubes, avoid hemolysis, aliquot, and freeze at -80 °C to limit degradation from repeated freeze-thaw cycles; clarify tissue homogenates by centrifugation at roughly 1000 x g for 10 minutes at 4 °C before loading. Cell culture supernatants may be assayed directly or concentrated by ultrafiltration when receptor levels are low. All reagents remain stable at 2-8 °C for the labelled shelf life when stored protected from light, and the 96-well microplate format readily accommodates duplicate or triplicate wells for standards, positive controls, and unknown samples on a single run.
