CTP synthase 1 (CTPS1) catalyzes the final step of de novo pyrimidine biosynthesis, converting UTP into CTP and supplying the nucleotide pools that fuel DNA replication, RNA transcription, and phospholipid turnover in actively dividing cells. This Assay Genie immunoassay is built on a microplate format and is calibrated to quantify human CTPS1 in serum or plasma-derived specimens using a two-antibody capture approach.

CTPS1 expression is a focal point in three active areas of research. In lymphocyte biology, the enzyme is essential for the rapid clonal expansion of T and B cells upon antigen stimulation, and partial loss-of-function variants have been identified as a cause of primary immunodeficiency in multiple kindreds. In oncology, elevated CTPS1 supports the nucleotide demand of proliferating tumor cells, and the enzyme is being evaluated as both a metabolic vulnerability and a candidate biomarker in lymphoid malignancies and selected solid tumors. Recent work has also extended CTPS1's role beyond nucleotide synthesis to the regulation of innate immune signaling pathways, including cGAS-STING and inflammasome activity.

The kit supplies pre-coated plates, capture and detection antibodies, a recombinant CTPS1 standard curve, and the buffers and enzyme-substrate reagents needed to perform the assay, and is intended for research use only. Reagents should be stored refrigerated, protected from light, and brought to room temperature before each run. Reconstituted standards should be aliquoted to limit freeze-thaw cycles and preserve binding activity through the stated shelf life.

The workflow follows standard plate immunoassay practice: dilute standards and samples in the provided buffer, add them to the wells, perform the sequential incubation and wash steps, develop the enzymatic reaction, and read absorbance in the wells. Standard curves generated on each plate enable interpolation of unknown samples, and the assay supports studies of nucleotide metabolism, immune cell proliferation, and tumor anabolism in both cell culture and ex vivo tissue work.
