Cytotoxic T-Lymphocyte-Associated Protein 4 (CTLA-4, also designated CD152) is a key negative regulator of T-cell activation that competes with CD28 for B7-1 and B7-2 binding on antigen-presenting cells. This Assay Genie microplate immunoassay enables quantitative measurement of soluble human CTLA-4, supporting research into tumor immunology, autoimmune disease, transplant tolerance, and immune checkpoint blockade therapies.

Using a matched antibody pair configuration on a pre-coated microplate, the assay captures CTLA-4 from research specimens, with a biotinylated detection antibody and streptavidin-HRP generating a colorimetric signal proportional to antigen concentration. The standard curve spans concentrations in the low pg/mL range, providing sensitivity sufficient to detect endogenous levels of soluble CTLA-4. Absorbance is read on a standard plate reader.

Each kit includes a pre-coated microplate, lyophilized recombinant CTLA-4 standards, detection antibody, streptavidin-HRP conjugate, wash buffer, and substrate solution, providing reagents for one full microplate run. Reconstituted standards should be aliquoted and stored frozen to limit freeze-thaw cycles. Reagents should be refrigerated where indicated, and light-sensitive components protected accordingly.

Validated for measurement of human CTLA-4 with no significant cross-reactivity against structurally related B7/CD28 family receptors at working concentrations, this research-use-only kit includes a comprehensive protocol covering standard preparation, sample handling, and data analysis guidelines.
