Description

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of human Creatine kinase B-type (CKB) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The kit detects natural and recombinant human CKB and is for research use only. CKB is a cytoplasmic enzyme that reversibly catalyses the transfer of phosphate between ATP and phosphogens such as creatine phosphate, playing a central role in energy transduction in tissues with large, fluctuating energy demands, including skeletal muscle, heart, brain and spermatozoa. Elevated levels of CKB have been associated with various cardiovascular and muscular diseases, making it a valuable biomarker in signal transduction research.

Reported performance specifications include a detection range of 0.156–10 ng/mL and a sensitivity of 0.078 ng/mL. Intra-assay CV is 4.6% and inter-assay CV is 7.8%. Linearity in serum (N=5) spans 103–113% at 1:2, 104–117% at 1:4, 105–114% at 1:8 and 97–107% at 1:16; EDTA plasma values are 99–109%, 84–93%, 107–117% and 90–102%; heparin plasma values are 104–114%, 104–115%, 101–113% and 101–111%. Recovery averages 85% (range 80–91) in serum and 87% (range 81–93) in plasma. CKB is a dimer of identical or non-identical B (brain) or M (muscle) chains, with MM predominant in skeletal muscle and myocardium, MB in myocardium, and BB in many tissues, especially brain. The molecular weight is 42,644 Da and the UniProt accession is P12277. Synonyms include brain creatine kinase, creatine kinase B chain, creatine phosphokinase B-type, B-CK, CPK-B and CKBB.

The assay is a colourimetric sandwich ELISA read at 450 nm. Other accepted sample types listed by the manufacturer include tissue homogenates, cell culture supernates and a range of biological fluids, alongside serum and plasma.
