Description

This sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of human CPA3 (Mast Cell Carboxypeptidase A), an enzyme primarily found in mast cells and involved in biological processes including inflammation and allergic responses. The assay uses a double-antibody sandwich format and is intended for use with serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. Elevated CPA3 levels have been associated with certain inflammatory conditions and allergic diseases, making the protein a valuable biomarker for related research.

The kit has a sensitivity of 0.469 ng/mL and a working range of 0.781–50 ng/mL, with reagents stored at 2–8°C for 12 months. Reported linearity in serum is 85–104% at 1:2, 85–102% at 1:4 and 91–104% at 1:8; in EDTA plasma 83–87% at 1:2, 90–98% at 1:4 and 85–101% at 1:8; and in heparin plasma 80–98% at 1:2, 80–94% at 1:4 and 82–100% at 1:8. Spike-and-recovery averages are 99% in serum (range 88–105%), 97% in EDTA plasma (range 86–104%) and 91% in heparin plasma (range 85–105%).

The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples, incubating at 37°C for 90 minutes for antigen binding, then sequential addition of a biotinylated detection antibody (37°C for 60 minutes), HRP-streptavidin (37°C for 30 minutes) and TMB substrate (10–20 minutes in the dark), with absorbance read at 450 nm after stop solution. Synonyms include MC-CPA and Carboxypeptidase A3 (UniProt P15088), and the intended research area is cell biology.
