Description

A Sandwich ELISA from Assay Genie designed for the quantitative measurement of C1qTNF3 (also referred to as CORS26, CTRP3, C1QTNF3, C1ATNF3, Cartducin, or cartonectin) in human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples [S1]. The assay uses a double-antibody sandwich format with colorimetric detection, a sensitivity of 0.188 ng/mL, and a measuring range of 0.313–20 ng/mL [S1].

Linearity has been validated in serum (n=5) at 83–97% (1:2), 85–100% (1:4), and 80–95% (1:8); in EDTA plasma (n=5) at 86–99%, 82–90%, and 87–100%; and in heparin plasma (n=5) at 84–100%, 87–101%, and 80–104% [S1]. Spike-and-recovery performance averages 97% for serum (n=5, range 85–101%), 95% for EDTA plasma (n=5, range 95–105%), and 93% for heparin plasma (n=5, range 85–101%) [S1]. The kit is intended for research use, including studies linking C1qTNF3 to inflammatory disorders, metabolic syndromes, and autoimmune diseases, and is stored at 2–8°C for up to 12 months [S1].

The assay procedure involves equilibrating reagents, a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody incubation, a 30-minute HRP-Streptavidin incubation, 10–20 minutes of TMB colour development in the dark, and absorbance reading at 450 nm after stop solution is added [S1]. Sample handling guidance includes clotting serum at room temperature and centrifugation at 1000 × g for 20 minutes, collecting plasma from anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2–8°C, and lysing cells or homogenising tissue in buffer with protease inhibitors prior to centrifugation [S1].
