Assay Genie's Human Copeptin (CPP) immunoassay quantitatively measures copeptin, the 39-amino-acid C-terminal glycopeptide cleaved from the precursor pro-arginine vasopressin during processing. Copeptin is co-secreted in equimolar amounts with mature vasopressin from the posterior pituitary but is far more stable in circulation, making it the preferred surrogate marker for vasopressin release in clinical research on chronic and acute heart failure, sepsis and septic shock, acute myocardial infarction, diabetes insipidus, and the differential diagnosis of polyuria-polydipsia syndromes.

The kit is configured on a standard microplate and is shipped with all reagents required to generate a standard curve and quantify endogenous copeptin in human samples, including serum-type preparations, plasma-type preparations (EDTA, heparin, citrate), urine, and cell culture supernatant. Capture antibody is pre-immobilised on the plate, and signal is generated through an enzyme-labelled detection antibody and chromogenic substrate solution readable on any benchtop plate reader capable of measuring absorbance in the visible range. Reconstituted standards, wash concentrate, and stop reagent are included. The standard curve is constructed from seven serial dilutions run in duplicate, with sample wells allocated alongside.

Reagent components should be stored refrigerated between uses to preserve antibody binding activity and standard curve integrity through the kit's stated shelf life. Reconstituted standards and working detector solution should be aliquoted to limit freeze-thaw cycles. The plate wash step uses the supplied wash concentrate diluted with deionised water, and incubations are typically carried out at room temperature protected from light. The kit is intended for research use only and is not validated for diagnostic procedures. Researchers commonly apply it to biomarker discovery, vasopressin axis studies, and evaluation of novel therapeutic interventions targeting the AVP pathway.
