Gap junction intercellular communication in cardiac muscle, neural tissue, and epithelial systems depends on Connexin 43 (CX43), the channel protein encoded by GJA1. Altered CX43 expression or phosphorylation is studied in the context of arrhythmia susceptibility, glial scar formation, wound re-epithelialization, and tumor cell bystander effects, making quantitative measurement of this phosphoprotein in tissue samples a recurring requirement in basic and translational research.

From Assay Genie, this ELISA-format kit is configured for human CX43 detection and provides a 96-assay microplate layout with pre-coated wells, calibrated recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, and TMB substrate solution. The colorimetric method is read on a standard microplate reader, and the validated workflow supports analysis of tissue homogenates and cell lysates prepared from cultured cells or snap-frozen surgical specimens.

For sample preparation, fresh or flash-frozen tissue should be homogenized in cold lysis buffer supplemented with protease and phosphatase inhibitors; adherent cell layers should be rinsed in ice-cold PBS before lysis. Clarify lysates by centrifugation, aliquot the supernatant into single-use tubes, and store at -80°C; limit freeze-thaw cycles to preserve the membrane-associated CX43 protein. Bring aliquots to room temperature immediately before the assay and protect working reagents from prolonged light exposure.

Intended for research use only, the kit is not validated for diagnostic or clinical decision-making and should be handled by trained laboratory personnel following standard biosafety practices.
