Human Connexin 40/GJA5 ELISA Kit

Assay Genie provides a sensitive sandwich ELISA built on a double-antibody format for the accurate quantification of Connexin 40 (GJA5) levels in human samples such as serum, plasma, and cell culture supernatants. The kit also accommodates cell or tissue lysate and other liquid samples, and uses a colorimetric detection method. Connexin 40 is a critical protein involved in cell-cell communication and plays a vital role in the proper functioning of the cardiovascular system, with dysregulation linked to various cardiac disorders and making it a valuable biomarker for studying these conditions.

The assay has a reported sensitivity of 0.188 ng/mL and a standard range of 0.313-20 ng/mL, and components should be stored at 2-8°C for 12 months. Recovery in serum (n=5) is reported at 85-98% with an average of 91%, in EDTA plasma (n=5) at 94-101% with an average of 97%, and in heparin plasma (n=5) at 85-102% with an average of 96%.

Linearity has been validated at dilution factors of 1:2, 1:4, and 1:8 across serum (93-103%, 85-101%, 82-96%), EDTA plasma (92-100%, 86-101%, 81-96%), and heparin plasma (91-103%, 84-101%, 87-99%). The procedure includes primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, and TMB substrate development followed by absorbance reading at 450 nm.
