Researchers studying cartilage and connective tissue biology can quantify Collagen Type IX Alpha 3 (COL9a3) using this Assay Genie ELISA kit, formatted for 96 assays in a microplate layout. Type IX collagen is a heterotrimeric FACIT-class extracellular matrix glycoprotein composed of three distinct alpha chains — COL9a1, COL9a2, and COL9a3 — encoded by separate genes. The COL9a3 polypeptide assembles with the other two chains to form a triple-helical molecule that decorates the surface of type II collagen fibrils in hyaline cartilage, the vitreous humor of the eye, and the tectorial membrane of the inner ear. Coding variants in COL9a3 have been linked to autosomal dominant multiple epiphyseal dysplasia type 3 (EDM3) and have been investigated as susceptibility factors for lumbar disc herniation and intervertebral disc disease. The immunoassay accommodates 96 reactions with absorbance-based readout and is intended for use with prepared human tissue homogenates, where a standard curve allows interpolation of COL9a3 concentrations. Prepared lysates should be refrigerated prior to running the assay. The kit supports investigations of osteoarthritis progression, age-related cartilage degeneration, and inherited connective tissue disorders.
