The assay is reactive with human samples and validated for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with specificity for natural and recombinant human Collagen alpha-1(XXVIII) chain.

Reported performance characteristics include a detection range of 0.156-10nmol/L, a sensitivity of 0.032nmol/L, an intra-assay CV of 5.2% and an inter-assay CV of 10.3%. Linearity was evaluated across dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma. Average recovery was 104% (range 98-110%) for serum and 106% (range 100-112%) for plasma. Optical density is read at 450 nm.

COL28A1 (UniProt Q2UY09) is a collagen family member described as potentially acting as a cell-binding protein, localised to the extracellular space, extracellular matrix and basement membrane as a 135 kDa monomer under reducing conditions or as a homotrimer under non-reducing conditions.
