Description

Assay Genie offers a Sandwich ELISA Kit designed for the quantitative measurement of human Collagen alpha-1 (X) chain (COL10A1). COL10A1 is a key component of collagen in the extracellular matrix, providing structural support to tissues such as bone and cartilage. Abnormalities in COL10A1 expression are associated with skeletal diseases such as chondrodysplasia and osteoarthritis, making it a useful biomarker for studying these conditions and developing potential therapies.

The assay is validated for use with serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids, and shows specificity for natural and recombinant human Collagen alpha-1(X) chain. Reported performance characteristics include a detection range of 0.156-10 ng/mL and a sensitivity of 0.098 ng/mL, with intra-assay precision of 6.2% CV and inter-assay precision of 11.2% CV. Recovery is reported as 114% on average for serum (range 108-120%) and 116% for plasma (range 110-120%). Linearity has been demonstrated across serum (96-116%), EDTA plasma (88-111%) and heparin plasma (89-118%) at dilutions of 1:2 to 1:16.

Type X collagen is encoded by the COL10A1 gene (NCBI Gene ID: 1300; UniProt Q03692), with a molecular weight of 66,158 Da. The protein is a homotrimer secreted to the extracellular space and extracellular matrix, and is annotated within the signal transduction research area. It is a product of hypertrophic chondrocytes, localised to presumptive mineralisation zones of hyaline cartilage, and mutations in COL10A1 are associated with Schmid type metaphyseal chondrodysplasia (SMCD) and Japanese type spondylometaphyseal dysplasia.
