Description

From Assay Genie, this sandwich ELISA is designed for the precise measurement of human Collagen alpha-1(X) chain (COL10A1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, using a colorimetric detection method.

Performance specifications include a detection range of 0.156-10 ng/mL and a sensitivity of 0.098 ng/mL. Intra-assay CV is 6.2% and inter-assay CV is 11.2%. Recovery averages 114% (range 108-120%) in serum and 116% (range 110-120%) in plasma. Linearity was assessed in serum, EDTA plasma and heparin plasma (each N=5) at dilutions of 1:2, 1:4, 1:8 and 1:16. Serum recovery was 96-106% at 1:2, 102-113% at 1:4, 102-114% at 1:8 and 105-116% at 1:16. EDTA plasma recovery was 97-107% at 1:2, 88-98% at 1:4, 95-103% at 1:8 and 101-111% at 1:16. Heparin plasma recovery was 89-102% at 1:2, 105-113% at 1:4, 108-118% at 1:8 and 110-118% at 1:16.

COL10A1 encodes the alpha chain of type X collagen, a short-chain collagen expressed by hypertrophic chondrocytes during endochondral ossification and localised to presumptive mineralisation zones of hyaline cartilage. The protein is a homotrimer secreted into the extracellular matrix, and abnormalities in its expression have been associated with skeletal diseases such as chondrodysplasia and osteoarthritis. Mutations in the gene are specifically linked with Schmid type metaphyseal chondrodysplasia (SMCD) and Japanese type spondylometaphyseal dysplasia (SMD). The assay shows specificity for natural and recombinant human Collagen alpha-1(X) chain and is intended for research use only.
