Human COL6/CollagenVI ELISA Kit

Assay Genie's sandwich ELISA kit is designed for the precise measurement of collagen VI levels in human samples. Using a double-antibody sandwich format with colorimetric detection, the kit is suited to serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Collagen VI is a vital component of the extracellular matrix, where it plays a key role in maintaining tissue integrity and providing structural support for cells; its dysregulation has been linked to muscular dystrophy, fibrosis, and skin disorders, making it a biomarker of interest in disease research and therapeutic development.

The assay has a sensitivity of 1.875 ng/mL and a working range of 3.125-200 ng/mL. The protocol involves equilibrating reagents and TMB substrate to room temperature, then incubating standards, samples, and blanks on the pre-coated plate at 37°C for 90 minutes. A biotin-labeled detection antibody is added for 60 minutes at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is added for 10–20 minutes in the dark, stop solution is applied, and absorbance is read at 450 nm. Unused strips should be stored at 2-8°C for 1 month or -20°C for 12 months.

Linearity was demonstrated in serum, EDTA plasma, and heparin plasma (n=5) at 1:2, 1:4, and 1:8 dilutions. Recovery ranges across sample types were 89-105% (serum, average 98%), 91-104% (EDTA plasma, average 99%), and 92-105% (heparin plasma, average 99%).
