Human COL4/CollagenIV ELISA Kit

Assay Genie supplies this sandwich ELISA for the quantitative measurement of human collagen IV (also known as COL4 or Type-IV Collagen) using a colorimetric, double-antibody detection method. The kit is intended for use with human samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are stored at 2-8°C for 12 months.

Reported performance characteristics include a sensitivity of 0.469 ng/mL and a working range of 0.781-50 ng/mL. Linearity was evaluated at 1:2, 1:4 and 1:8 dilutions across serum (87-105%, 83-98%, 84-100%), EDTA plasma (86-100%, 86-97%, 85-100%) and heparin plasma (86-98%, 82-101%, 80-100%), with n=5 per matrix. Spike-and-recovery averages were 98% for serum, 98% for EDTA plasma and 90% for heparin plasma, with corresponding ranges of 89-103%, 92-105% and 86-103%.

Collagen IV is a key structural component of the extracellular matrix, supporting tissue integrity and contributing to cell adhesion, migration and differentiation. Abnormal collagen IV levels have been linked to kidney diseases, vascular disorders and certain cancers, making it a useful biomarker in related research. The Assay Genie protocol follows six steps: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB colour development in the dark for 10-20 minutes, and a stop reaction read at 450 nm.
