Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of human COL17A1 (Collagen XVII) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, using a double-antibody sandwich format with colorimetric detection. It is intended for research applications in dermatology and related fields.

Collagen XVII, also known by the synonyms COL17, BP180, BPAG2, and 180 kDa bullous pemphigoid antigen 2, is a transmembrane collagen associated with dermal-epidermal adhesion; its dysregulation has been linked to skin disorders including bullous pemphigoid. The assay provides a sensitivity of 1.875 ng/mL and a measurable range of 3.125-200 ng/mL. Linearity in serum (n=5) at dilutions of 1:2, 1:4, and 1:8 is reported at 90-105%, 84-93%, and 88-94%, with EDTA plasma at 85-103%, 87-100%, and 83-100%, and heparin plasma at 91-101%, 84-100%, and 81-95%. Recovery averages 95% for serum, 95% for EDTA plasma, and 97% for heparin plasma.

The protocol comprises six steps: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Reagents should be stored at 2-8°C for 12 months.
