The Human CLEC10A/CD301 ELISA Kit from Assay Genie provides quantitative measurement of CLEC10A, a type II transmembrane glycoprotein in the C-type lectin superfamily also known as macrophage galactose-type lectin (MGL) or CD301. CLEC10A is expressed predominantly on alternatively activated macrophages and selected dendritic cell subsets, where it recognizes terminal N-acetylgalactosamine residues and the tumor-associated Tn antigen on glycoproteins and pathogen surfaces.

Researchers use this kit in studies of tumor immune evasion, antigen uptake by tolerogenic myeloid cells, M2 macrophage polarization, and glycan-dependent immune regulation in infectious or inflammatory disease models. Capture antibody is pre-bound on the plate, bound analyte is detected with a biotinylated secondary antibody followed by streptavidin-HRP and a chromogenic substrate, and a recombinant human CLEC10A standard is included for quantitative calibration. The recommended sample volume is 50 uL per well, loaded alongside serially diluted standards in duplicate.

For reliable results, serum or plasma samples should be aliquoted promptly and stored frozen at -20 degrees C to limit freeze-thaw cycles, with working aliquots kept refrigerated until use. Kit reagents should equilibrate to room temperature before each step. After incubation, the plate is washed, substrate is added, and the reaction is stopped before reading absorbance. The complete kit is supplied as a single pack containing a pre-coated microplate, detection reagents, wash buffer, substrate, and stop solution, intended for research use only.
