Human CLDN9/Claudin-9 ELISA Kit

Manufactured by Assay Genie, this sandwich ELISA kit is designed for the precise measurement of CLDN9 levels in human serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The kit uses a colorimetric detection method in a double antibody sandwich format and offers a sensitivity of 46.875 pg/mL with a measuring range of 78.125-5000 pg/mL. CLDN9 is a vital protein involved in cell-cell adhesion and the maintenance of epithelial barrier function, and its dysregulation has been linked to inflammatory bowel disease, hepatitis C, and cancer, making it a valuable biomarker for studying these conditions and developing potential therapeutic interventions.

The assay procedure begins with equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, and binding antigens at 37°C for 90 minutes. A biotin-labeled detection antibody is then added with a 60-minute incubation at 37°C, followed by HRP-Streptavidin binding for 30 minutes. Color is developed with TMB substrate in the dark for 10-20 minutes, and absorbance is read at 450 nm immediately after adding stop solution.

Validated linearity across dilution factors of 1:2, 1:4 and 1:8 produced values of 86-105%, 84-98% and 82-97% in serum (n=5); 92-97%, 87-101% and 82-88% in EDTA plasma (n=5); and 86-97%, 84-95% and 81-100% in heparin plasma (n=5). Recovery rates were 87-103% in serum (average 94%), 88-102% in EDTA plasma (average 95%) and 87-101% in heparin plasma (average 94%). The kit should be stored at 2-8°C for 12 months.
