Assay Genie's CLDN4 chemiluminescence immunoassay is built on a sandwich-type antibody pair immobilized on a 96-well microplate, capturing human claudin-4 directly from biological specimens. Each kit ships with a pre-coated microwell plate, lyophilized recombinant CLDN4 standards for curve generation, biotinylated detection antibody, streptavidin-HRP conjugate, chemiluminescent substrate solution, concentrated wash buffer, and assay diluent. A typical workflow loads 50-100 µL of sample or standard per well, incubates the plate overnight at 2-8 °C to allow antigen binding, then adds detection reagent for a 60-minute incubation at room temperature, followed by conjugate binding, washing, and signal development. Luminescent output is read on a luminometer or glow-capable microplate reader, with claudin-4 concentration interpolated from the standard curve. Because claudin-4 is implicated in epithelial-mesenchymal transition and is frequently overexpressed in peritoneal dissemination models, the assay is commonly adopted for biomarker validation comparing tumor and matched normal tissue lysates, ascites fluid, and circulating exosome preparations. Researchers should equilibrate all reagents to room temperature before use, avoid cross-contamination by using fresh discarded tips, and run samples in duplicate or triplicate to minimize intra-assay variability. Reconstituted standards should be aliquoted and stored at -20 °C or below to preserve immunoreactivity across repeated freeze-thaw cycles. Full assay time generally fits within a single working day, and the kit is calibrated for research use only in human in vitro diagnostic development and translational oncology workflows.
