Human CK13/KRT13/Cytokeratin 13 ELISA Kit

This sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of human cytokeratin 13 (KRT13) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Using a double-antibody sandwich format with colorimetric detection, the kit provides a sensitivity of 9.375 pg/mL and a quantitative range of 15.625-1000 pg/mL. Cytokeratin 13 is a key protein in the network of intermediate filaments that make up the cytoskeleton of epithelial cells, where it plays a crucial role in maintaining the structural integrity and function of epithelial tissues, making it an important biomarker for studying epithelial-related diseases such as cancer, skin disorders, and inflammatory conditions.

The assay protocol includes equilibrating reagents and TMB substrate to room temperature, then loading standards, samples and controls onto the pre-coated plate for a primary incubation of 90 minutes at 37°C. A biotin-labeled detection antibody is added for 60 minutes, then HRP-Streptavidin conjugate (SABC) for 30 minutes, before TMB substrate is added for 10-20 minutes in the dark. After the stop solution is added, absorbance is read at 450 nm. Sample preparation guidelines are included for serum, plasma, tissue homogenate, cell culture supernatant and cell lysate.

Validation data demonstrate linearity for serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5) across serial dilutions of 1:2, 1:4 and 1:8, with recovery ranging from 95-103% (serum), 89-100% (EDTA plasma) and 91-105% (heparin plasma). The kit is stored at 2-8°C for 12 months and is supplied with a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody, HRP-Streptavidin conjugate, TMB substrate, sample, antibody and SABC dilution buffers, stop solution, wash buffer, plate sealer and technical manual.
