Human CFLAR/CASP8 and FADD-like apoptosis regulator ELISA Kit

A sandwich ELISA, double antibody assay from Assay Genie designed for the quantitative detection of human CFLAR (CASP8 and FADD-like apoptosis regulator). The colorimetric kit is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a reported sensitivity of 46.875 pg/mL and a standard curve range of 78.125–5000 pg/mL.

CFLAR is a key regulator of apoptosis, and its dysregulation has been implicated in cancer, autoimmune disorders, and neurodegenerative conditions. Reported linearity values are: serum (n=5) 87–93% at 1:2, 85–105% at 1:4, 88–102% at 1:8; EDTA plasma (n=5) 85–101%, 82–98%, and 85–94%; heparin plasma (n=5) 80–93%, 82–100%, and 84–98%. Recovery ranges are: serum 93–100% (average 91%), EDTA plasma 92–100% (average 96%), and heparin plasma 86–99% (average 96%). Reagents are stored at 2–8°C for 12 months.

The workflow includes reagent and TMB equilibration, primary incubation at 37°C for 90 minutes, biotin-labelled antibody binding for 60 minutes at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB color development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Kit components include a dismountable microplate, lyophilised standard, 100X biotin-labelled antibody, 100X HRP-streptavidin conjugate, TMB substrate, sample/antibody/SABC dilution buffers, stop solution, 25X wash buffer, plate sealer, and a technical manual.
