Description

Designed by Assay Genie, this Sandwich ELISA Kit uses a double-antibody, colorimetric method to measure CDKN2A (p16INK4a) levels in human serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The kit is intended for research applications, including the study of CDKN2A, a tumour suppressor gene involved in cell cycle regulation that has been associated with cancers such as melanoma and pancreatic cancer.

Reported sensitivity is 0.375 ng/mL with a standard range of 0.625-40 ng/mL. Performance data for serum, EDTA plasma and heparin plasma show linearity between 82% and 105% across 1:2, 1:4 and 1:8 dilutions (n = 5), and average recovery values of 96% for serum, 96% for EDTA plasma and 92% for heparin plasma. Components include a dismountable ELISA microplate, lyophilised standard, biotin-labelled detection antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer and plate sealer, with storage at 2-8°C for 12 months.

The assay procedure involves equilibrating reagents and the TMB substrate to room temperature, incubating the plate at 37°C for 90 minutes for primary antigen binding, adding biotin-labelled detection antibody for 60 minutes at 37°C, incubating with HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing colour with TMB substrate for 10-20 minutes in the dark, adding stop solution and reading absorbance at 450 nm. Associated UniProt annotation: P42771.
