Human Cdk4 ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of Cyclin-Dependent Kinase 4 (CDK4) in human samples including serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. CDK4 is a key regulatory protein involved in cell cycle progression, particularly the G1/S transition, and its dysregulation has been linked to various diseases such as cancer, making it an important biomarker for studying tumorigenesis and disease pathogenesis.

The assay uses a double-antibody sandwich format with colorimetric detection. Reported sensitivity is 0.188 ng/mL and the working range is 0.313–20 ng/mL. Kit components should be stored at 2–8°C for 12 months. The protocol involves equilibrating reagents and the TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after addition of stop solution.

Validation data include linearity and recovery testing across serum, EDTA plasma, and heparin plasma. Recovery ranges were 89–101% (average 94%) for serum, 86–105% (average 96%) for EDTA plasma, and 85–105% (average 97%) for heparin plasma. Linearity at 1:2, 1:4, and 1:8 dilutions was 92–95%, 80–100%, and 86–101% for serum; 89–104%, 92–95%, and 80–100% for EDTA plasma; and 86–101%, 83–100%, and 80–95% for heparin plasma.
