Description

A sandwich ELISA kit from Assay Genie designed for the quantitative detection of human CD69 (also known as CLEC2C), a cell surface glycoprotein involved in lymphocyte proliferation and signal transduction in T cells, NK cells, and platelets. The assay uses a colorimetric detection method and is intended for research use only.

According to the manufacturer's product listing, the kit is reactive with human samples and has been validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported analytical performance includes a sensitivity of 46.875 pg/mL, a working range of 78.125–5000 pg/mL, and recovery values for serum (average 100%), EDTA plasma (average 96%) and heparin plasma (average 98%). Spiked linearity across 1:2, 1:4 and 1:8 dilutions is reported as 86–99% / 82–100% / 84–99% for serum, 91–105% / 83–97% / 80–94% for EDTA plasma and 85–95% / 83–100% / 80–96% for heparin plasma (n = 5 for each).

The assay protocol is a sandwich format using a biotin-labelled detection antibody followed by HRP-Streptavidin (SABC) and TMB substrate, with absorbance read at 450 nm. Key incubation steps include 90 minutes at 37°C for primary antigen binding, 60 minutes at 37°C for detection antibody binding and 30 minutes at 37°C for HRP-Streptavidin binding, with TMB colour development performed in the dark for 10–20 minutes. The kit should be stored at 2–8°C for 12 months.
