CD40L, also known as CD154 or TNFSF5, is a type II transmembrane glycoprotein of the TNF superfamily that plays a central role in adaptive immunity by engaging CD40 on B cells, dendritic cells, macrophages, and endothelial cells. This binding drives B-cell proliferation, immunoglobulin class-switch recombination, germinal center formation, and pro-inflammatory cytokine release, placing CD40L at the center of research into autoimmunity, transplant biology, vaccination response, atherosclerosis, and platelet-mediated inflammation.

The Assay Genie CD40L ELISA kit provides an in vitro quantitative immunoassay for measuring human CD40L in cell culture supernatant, plasma, and serum samples, supporting immunology, inflammation, and biomarker discovery workflows. The kit is configured for 96 assays in a microplate layout, with reagents prepared for standard curve generation and direct sample measurement. Intended for research use only, it supports reproducible quantification of soluble ligand released from activated T cells and platelets.

Relevant applications include profiling T-cell costimulatory activity, evaluating B-cell helper function in cell culture, characterizing immune regulation in autoimmune disease models, and tracking soluble CD40L as a circulating biomarker in translational studies. Investigators focused on hyper-IgM syndrome mechanisms, allograft rejection monitoring, or thrombotic and inflammatory pathways will find this assay useful for quantifying ligand release under varied stimulation conditions.
