Human CD3e/CD3 epsilon ELISA Kit

A Sandwich ELISA kit from Assay Genie for the quantitative measurement of human CD3e/CD3 epsilon in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a double-antibody format with colorimetric detection and is intended for immunology research. CD3e is a component of the T-cell receptor complex involved in T-cell activation and immune responses, and is implicated in autoimmune and immunodeficiency conditions.

The assay reports a sensitivity of 0.375 ng/mL and a standard range of 0.625-40 ng/mL. Linearity assessed at 1:2, 1:4, and 1:8 dilutions (n=5) was 86-105%, 88-101%, and 83-100% for serum; 96-105%, 83-96%, and 81-94% for EDTA plasma; and 85-103%, 83-101%, and 80-92% for heparin plasma. Spike-and-recovery performance showed ranges of 87-105% (average 95%) for serum, 90-103% (average 95%) for EDTA plasma, and 90-105% (average 97%) for heparin plasma.

The procedure includes equilibrating reagents and TMB substrate to room temperature, incubating standards and samples on the pre-coated plate at 37°C for 90 minutes, adding biotin-labeled detection antibody and incubating at 37°C for 60 minutes, adding HRP-Streptavidin (SABC) and incubating at 37°C for 30 minutes, developing with TMB substrate for 10-20 minutes in the dark, and adding stop solution before measuring absorbance at 450 nm. Serum is clotted and centrifuged at 1000 × g for 20 minutes; plasma is collected with anticoagulant and centrifuged at 1000 × g for 15 minutes at 2-8°C; cell culture supernatant is centrifuged at 2500 rpm for 5 minutes. Kit components are stored at 2-8°C for 12 months.
