Human CD28 ELISA Kit

Assay Genie's Human CD28 ELISA Kit is a Sandwich ELISA (Double Antibody) using a colorimetric detection method to quantify CD28 levels in human samples. Intended for cell biology research, the kit is suited to studies of T-cell co-stimulation, immune response modulation, autoimmune disease, transplant rejection and inflammatory conditions. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The kit is reported to offer a sensitivity of 0.375 ng/mL and a measuring range of 0.625-40 ng/mL. Reported linearity (n=5) across 1:2, 1:4 and 1:8 dilutions is 87-100%, 93-105% and 88-102% for serum; 84-100%, 82-95% and 84-99% for EDTA plasma; and 80-97%, 82-97% and 81-94% for heparin plasma. Spike-and-recovery averages (n=5) are 97% for serum (range 90-103%), 93% for EDTA plasma (range 87-99%) and 99% for heparin plasma (range 89-104%). Unopened kit components are stored at 2-8°C for up to 12 months.

The assay workflow involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and a stop reaction with absorbance measurement at 450 nm.
