Description

This sandwich double-antibody ELISA kit from Assay Genie is designed for the quantitative measurement of human CD163 across a broad working range of 15.625–1000 pg/mL, with a reported sensitivity of 9.375 pg/mL. The assay is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and the microplate is supplied pre-coated with a capture antibody specific to the target protein.

Samples, standards and a zero control are loaded onto the plate and incubated at 37 °C for 90 minutes to allow antigen binding. A biotin-labelled detection antibody directed against a separate epitope is then added and incubated for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is added and developed in the dark for 10–20 minutes; the reaction is stopped and absorbance is read immediately at 450 nm, with signal intensity proportional to CD163 concentration as determined from a standard curve. Sample preparation guidance covers serum (clot, 1000 × g, 20 min), plasma (anticoagulant tubes, 1000 × g, 15 min, 2–8 °C), tissue homogenate, cell culture supernatant (2500 rpm, 5 min) and cell lysate protocols. Reported linearity for dilutions of 1:2, 1:4 and 1:8 spans approximately 80–103% across serum, EDTA plasma and heparin plasma, with recovery averages of 94% (serum), 99% (EDTA plasma) and 94% (heparin plasma). Unused components should be stored at 2–8 °C, with lyophilised standard and plate strips stable for 12 months at –20 °C; the kit itself is stored at 2–8 °C for 12 months.
