Description

Assay Genie's sandwich ELISA kit is designed for the quantitative measurement of human C-C chemokine receptor type 7 (CCR7) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Built around a double-antibody sandwich format with colorimetric detection, the assay targets CCR7 (UniProt P32248), a protein implicated in immune cell trafficking. Because dysregulation of CCR7 has been associated with inflammatory diseases, autoimmune disorders and cancer progression, the kit is positioned for research into immune system function, inflammation and related conditions.

Reported linearity for serum (n=5) spans 85–105% at 1:2, 91–105% at 1:4 and 86–101% at 1:8, with EDTA plasma at 82–100%, 82–97% and 83–100% and heparin plasma at 84–99%, 87–98% and 84–100% across the same dilutions. Recovery averages are 91% for serum, 92% for EDTA plasma and 95% for heparin plasma, with corresponding recovery ranges of 86–96%, 86–102% and 88–102%.

Storage is at 2–8°C for 12 months. The protocol involves primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding for 30 minutes at 37°C, TMB colour development in the dark for 10–20 minutes and a stop step with absorbance read at 450 nm. Sample preparation guidance covers clotting serum at 1000 × g for 20 minutes, anticoagulant plasma at 1000 × g for 15 minutes at 2–8°C, tissue homogenisation in PBS with protease inhibitors, cell culture supernatant clarification at 2500 rpm for 5 minutes and cell lysis with protease inhibitors.
