The Assay Genie Human CC16 ELISA Kit uses a pre-coated 96-well microplate in sandwich configuration, with a biotinylated detection antibody paired with streptavidin-HRP for colorimetric signal generation. The assay typically demonstrates a dynamic range of approximately 0.156–10 ng/mL, with intra-assay and inter-assay precision generally below 10% CV, supporting reproducible quantitation in serum, plasma, bronchoalveolar lavage fluid, urine, and cell culture supernatants.

Each kit contains the pre-coated plate, recombinant human CC16 standards, biotinylated detection antibody, streptavidin-HRP conjugate, concentrated wash buffer, TMB substrate, stop solution, and protocol booklet. The workflow involves adding standards and samples to coated wells, incubation, washing, detection antibody addition, a second incubation and wash, TMB development protected from light, stop solution, and absorbance measurement at 450 nm with a 570 nm or 630 nm reference wavelength on a standard microplate reader. Reagents should be stored refrigerated at 2–8 °C; standards are best aliquoted and frozen at -20 °C to limit freeze-thaw cycles, and the kit is shipped on ice packs with a shelf life of several months when stored as directed.

Beyond the indications already described, CC16 has been investigated as a biomarker of inhaled toxicant exposure, silica and particulate matter injury, electronic cigarette aerosol effects, and chronic kidney disease–associated reductions in renal clearance. For research use only; not for diagnostic procedures.
