Description
A colorimetric Sandwich ELISA from Assay Genie designed for the quantitative measurement of human Caspase-1, an enzyme central to inflammatory processes and cell death pathways. The kit shows high sensitivity and specificity, with detection in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 46.875 pg/mL across a standard range of 78.125–5000 pg/mL, and reagents should be stored at 2–8°C for up to 12 months.
Validated linearity is demonstrated at 1:2, 1:4 and 1:8 dilutions for serum (90–102%, 89–101%, 91–104%), EDTA plasma (82–99%, 82–100%, 84–98%) and heparin plasma (81–99%, 80–92%, 82–100%). Recovery averages 96% for serum, 95% for EDTA plasma and 92% for heparin plasma. The assay is intended for research use, including studies of inflammatory, autoimmune and neurodegenerative disease contexts where abnormal Caspase-1 activation is of interest.
The protocol follows a standard sandwich workflow: plate and reagent equilibration, primary incubation at 37°C for 90 minutes, biotinylated detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after stop solution addition. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant and cell lysate, with technical support available for additional sample types.
