Caspase 7 is encoded by the CASP7 gene on chromosome 10q25.3 and is synthesized as a 35 kDa inactive zymogen that becomes activated through proteolytic cleavage at Asp198 and Asp206, yielding the p20 and p12 subunits characteristic of executioner caspases. As an executioner protease, CASP7 shares the DEVD tetrapeptide substrate preference with caspase-3 but exhibits distinct cleavage kinetics toward certain substrates including PARP, lamin A, and DNA fragmentation factor 40. This Sandwich ELISA Kit from Assay Genie uses pre-coated monoclonal capture antibodies on a 96-well microplate, with bound CASP7 detected via an HRP-conjugated secondary antibody and TMB substrate; the enzymatic reaction is terminated with sulfuric acid stop solution and absorbance is read at 450 nm. Typical standard curves span 0.156–10 ng/mL, with intra-assay CV under 10% and inter-assay CV under 12%, enabling quantitative comparison across biological replicates. The protocol accepts both fresh and frozen aliquots prepared in RIPA or comparable lysis buffer, though repeated freeze-thaw cycles should be minimized to preserve epitope integrity. Beyond apoptosis and oncology work, the kit has been applied to chemotherapy sensitivity screening, ischemic injury models, and neurodegeneration studies evaluating mitochondrial pathway involvement. Researchers commonly pair this CASP7 measurement with parallel caspase-3, caspase-9, and PARP cleavage readouts to distinguish intrinsic versus extrinsic apoptotic signaling in the same experimental cohort. All reagents are shipped refrigerated and should remain stored at 2-8 °C until first use, with the detection antibody and TMB working solution kept away from light. For research use only, the kit is not intended for diagnostic procedures or clinical patient testing.
