Description

A Sandwich ELISA designed by Assay Genie for the quantitative measurement of Carboxypeptidase M (CPM) in human samples. The kit is configured for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and uses a colorimetric, double-antibody sandwich detection format with absorbance read at 450 nm.

Reported assay performance includes a sensitivity of 0.938 ng/mL and a measurement range of 1.563–100 ng/mL. Linearity in spiked serum (n=5) is 82–95% at 1:2, 83–97% at 1:4, and 80–99% at 1:8; EDTA plasma (n=5) shows 83–95%, 87–105%, and 85–100% at the same dilutions, while heparin plasma (n=5) shows 82–101%, 87–96%, and 85–94%. Spike-and-recovery averages are 97% across serum (range 92–100%), EDTA plasma (range 85–103%), and heparin plasma (range 92–99%). Components should be stored at 2–8°C for 12 months, with standards and coated strips also suitable for longer-term storage at –20°C.

The protocol runs as a six-step procedure: equilibrate reagents and TMB substrate, primary incubation at 37°C for 90 minutes, biotinylated detection antibody for 60 minutes, HRP-Streptavidin for 30 minutes, TMB color development in the dark for 10–20 minutes, and stop solution followed by immediate absorbance reading. Sample-specific preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate. The kit is intended for cell biology research, with CPM also referenced by the synonym Carboxypeptidase M and Uniprot ID P14384.
