Human Carbonic Anhydrase VB/CA5B ELISA Kit

A high-quality sandwich ELISA from Assay Genie designed for the quantitative detection of human Carbonic Anhydrase VB (CA5B), a mitochondrial enzyme. The double-antibody sandwich format is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Carbonic anhydrase VB plays a role in pH regulation and is involved in processes such as bone resorption and calcification; alterations in CA5B levels have been linked to osteoporosis, kidney stones, and certain cancers, making accurate measurement valuable for related research.

Linearity was assessed by serial dilution of spiked samples: serum (n=5) 86–104% at 1:2, 85–101% at 1:4, 88–105% at 1:8; EDTA plasma (n=5) 85–101%, 82–100%, 86–100%; heparin plasma (n=5) 82–100%, 89–98%, 81–98%. Spike-and-recovery testing yielded serum (n=5) 85–105% with a 94% average, EDTA plasma (n=5) 95–104% with a 99% average, and heparin plasma (n=5) 85–99% with a 93% average.

The workflow involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks for a 37°C primary incubation of 90 minutes, adding a biotin-labeled detection antibody (37°C for 60 minutes), adding HRP-Streptavidin (37°C for 30 minutes), developing colour with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm immediately after adding stop solution.
