This Assay Genie ELISA kit enables quantitative measurement of carbonic anhydrase IV (CA4) in research samples, with applications spanning renal and ocular physiology. CA4 is a glycosylphosphatidylinositol (GPI)-anchored zinc metalloenzyme expressed on the apical plasma membrane of renal proximal tubule cells and in corneal endothelium, where it accelerates the reversible hydration of carbon dioxide. Loss-of-function variants in the CA4 gene have been linked to autosomal dominant retinitis pigmentosa (RP17), positioning CA4 as a target of interest in studies of acid-base balance, carbonic anhydrase inhibitor pharmacology, and hereditary retinal disease.

Each kit is supplied as a complete reagent set sized for 96 individual determinations and works with human-derived liquid samples and tissue preparations. Reagents should be stored refrigerated at 4°C; allow the kit to equilibrate to room temperature before running. The procedure uses a standard curve-based assay workflow with detailed plate layout, standard dilution series, and assay buffer preparation provided in the protocol insert. Reconstituted standards and detection antibody should be aliquoted to limit freeze-thaw cycles and preserve signal stability across runs.

Typical applications include biomarker studies in nephrology, ophthalmic research, and characterization of patient-derived material in hereditary retinal dystrophy. The kit is intended for research use only and is not validated for clinical diagnostic procedures.
