Description

This Sandwich ELISA from Assay Genie is intended for the quantitative detection of Human Carbohydrate Antigen 50 (CA50) in human samples, with use supported across serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. The assay uses a double-antibody sandwich configuration with a colorimetric readout, and the kit provides a pre-coated microplate, lyophilised standard, biotinylated detection antibody, HRP-Streptavidin conjugate, TMB substrate, and the dilution and wash buffers required to perform the test.

CA50 is a carbohydrate antigen commonly linked to cancers such as pancreatic, colorectal, and gastric cancer, and monitoring CA50 levels can support research into cancer biomarkers, disease progression, and treatment effectiveness. The assay reports a sensitivity of 18.75 pg/mL and a detection range of 31.25-2000 pg/mL (2000pg=3.2U). Linearity has been confirmed in serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, and recovery across these sample types ranges from 85% to 104%. Reagents are stored at 2-8°C and remain stable for 12 months under those conditions.

The run protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples for a 90-minute primary incubation at 37°C, binding the biotin-labelled detection antibody for 60 minutes at 37°C, incubating with HRP-Streptavidin for 30 minutes at 37°C, developing colour with TMB substrate in the dark for 10–20 minutes, adding stop solution, and reading absorbance at 450 nm.
