Human CAPN2/Calpain 2 ELISA Kit

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of human CAPN2 (Calpain-2), a calcium-dependent protease implicated in cell migration, adhesion, and apoptosis. The kit can be used with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Dysregulation of calpain-2 has been linked to muscular dystrophy, neurodegenerative disorders, and cancer, making this assay a useful tool for biomarker and therapeutic research. Detection is colorimetric using a double-antibody sandwich format.

Linearity in serum at 1:2, 1:4, and 1:8 dilutions is 87–105%, 89–101%, and 82–99% respectively; in EDTA plasma, 85–97%, 84–89%, and 82–100%; and in heparin plasma, 88–102%, 98–100%, and 83–98%. Recovery in serum averages 91% (range 87–101%), 96% in EDTA plasma (range 92–105%), and 96% in heparin plasma (range 86–104%). The assay protocol involves a 90-minute primary incubation at 37°C, biotinylated detection antibody binding for 60 minutes, HRP-Streptavidin binding for 30 minutes, and TMB substrate development in the dark for 10–20 minutes, with absorbance read at 450 nm after stop solution is added. Unopened kits are stored at 2–8°C for 12 months.
